I prefer to receive a short letter/note, on an A4 page (it's easier to file than an odd shaped card - now we're getting into details!).
Just very brief and to the point - I already know everything about the applicant, I don't need to know anymore, or waste time re-reading things. It's the pro-activeness of the applicant that is remembered when a thank you letter is sent, not the content of the actual letter.
Oh, and don't forget the thank you card if you do get the job - in this case a card is more appropriate because it can be displayed.

Great advice. In fact, in the past I have always used a short letter on an A4 page.
