Hi,
I am a frustrated PhD who wants to relocate myself and family to the RTP area in NC. I have applied to several jobs, however, I haven't landed any interviews. Is it acceptable to make calls to big companies or is that frowned upon? I am coming from the academia world to industry and I just can't figure out the system. Does the squeaky wheel get the grease or will they throw my resume away if I call to follow up on jobs that I've applied for?

If you make cold calls (it isn't clear what you mean, but I assuming you mean calls via telephone), all that will happen is you will leave messages on voice mail, and you won't hear back.
If you just send a resume, there's a slight (very, very) slight chance you might get lucky, but more likely what will happen is that you won't hear anything back, and if you decide to get professional help to do it right (you DID say that you "just can't figure out the system"), you will have already "poisoned the well" - meaning, you usually only get one chance at applying for a job (I realize you didn't say anything about actually applying for a position, but I assume you would try to give them at least SOME idea about what you want. Otherwise, you really would be wasting your time/energy and STILL be poisoning the well).
The hirng process is largely one of elimination, NOT selection. One of the most important things to know about "the system" is that co's use resumes to screen people out, and it takes less than a nanosecond to see if your current title is similar to an opening they have, and if it is, do you have current experience in THEIR industry.
Furthermore, from their perspective, unless you have some critically-needed skills they can't readily find in "RTP" (whatever that is), they are NOT going to even bother paying for you to come and interview, much less pay to move your family there.
Assuming that you are serious, you really should get professional help. It isn't cheap, but nothing good is. (how much did getting your PhD cost? a lot, I know, because I have a cousin that has one)
