I would suggest that you approach this head on.
"Boss, I am aware that you spent considerable time and money hiring me, but I have to be honest: I am not happy here and feel that it is in everyone's best intrest that I tender my resignation."
I suspect that he/she will most likely appreciate the directness. Those canidates who came in second or third place may still be available. It may not involve much trouble or expense to refill your position.
Also:
I would not mention the new position if you can possibly avoid doing so.

I am going through the same situation. I just joined a firm about a month ago and thought this job was going to be much more outward, sales-facing. So far I have crunched spreadsheets for the last three weeks and done policy statements. Pretty dull stuff when I expected lots of work with Sales.
My boss knows I am unhappy, and I don't have to say anything. He just looks at me like "I'm sorry..."
So chances are your boss knows already and can sense it. Just lay it out and don't ever talk about the new deal. Just say you are resiging as you respect the organization's goals and know long-term you are not the right person for the job. Quick, clean, direct - just have all your stuff packed before you say those words!
Be strong and deliver it straight up, they will respect you for it.
